Single-cell transcriptomics of the Drosophila wing disc reveals instructive epithelium-to-myoblast interactions

چکیده

In both vertebrates and invertebrates, generating a functional appendage requires interactions between ectoderm-derived epithelia mesoderm-derived cells. To investigate such interactions, we used single-cell transcriptomics to generate temporal cell atlas of the Drosophila wing disc from two developmental time points. Using these data, visualized gene expression using multilayered model cataloged ligand–receptor pairs that could mediate signaling epithelial cells adult muscle precursors (AMPs). We found localized fibroblast growth factor ligands, Thisbe Pyramus, in epithelium regulates number location AMPs. addition, Hedgehog ligand activates specific transcriptional program within adjacent AMP cells, defined by AMP-specific targets Neurotactin midline , is critical for proper formation direct flight muscles. More generally, our annotated provides an organ-wide view potential cell–cell myogenic

CDC42 and Rac1 control different actin-dependent processes in the Drosophila wing disc epithelium

Cdc42 and Rac1 are members of the rho family of small guanosinetriphosphatases and are required for a diverse set of cytoskeleton-membrane interactions in different cell types. Here we show that these two proteins contribute differently to the organization of epithelial cells in the Drosophila wing imaginal disc. Drac1 is required to assemble actin at adherens junctions. Failure of adherens jun...

Actin capping protein alpha maintains vestigial-expressing cells within the Drosophila wing disc epithelium.

Tissue patterning must be translated into morphogenesis through cell shape changes mediated by remodeling of the actin cytoskeleton. We have found that Capping protein alpha (Cpa) and Capping protein beta (Cpb), which prevent extension of the barbed ends of actin filaments, are specifically required in the wing blade primordium of the Drosophila wing disc. cpa or cpb mutant cells in this region...

Single cell transcriptomics reveals unanticipated features of early hematopoietic precursors

Molecular changes underlying stem cell differentiation are of fundamental interest. scRNA-seq on murine hematopoietic stem cells (HSC) and their progeny MPP1 separated the cells into 3 main clusters with distinct features: active, quiescent, and an un-characterized cluster. Induction of anemia resulted in mobilization of the quiescent to the active cluster and of the early to later stage of cel...

Comparative Generalized Logic Modeling Reveals Differential Gene Interactions during Cell Cycle Exit in Drosophila Wing Development

A comparative interaction detection paradigm is proposed to study the complex gene regulatory networks that control cell proliferation during development. Instead of attempting to reconstruct the entire cell cycle regulatory network from temporal transcript data, differential interactions - represented by generalized logic - are detected directly from time course transcript data under two disti...
